Chromatin landscape and regulatory networks are determinant in cell lineage specification differentiation. We generated profiles of expression signature (RNA-Seq), chromatin accessibility (ATAC-Seq), architecture (Hi-C), histone modifications (ChIP-Seq) neonate epidermis vivo to define the modules governing murine skin observed that 80% differentially regulated genes repressed differentiated epidermal fraction, with super-enhancers (SEs) controlling differentiation-specific epigenomic changes. The results underscore relevance Dlx/Klf/Grhl combinatorial network maintaining precise gene during determined differential open compartments, topologically associating domain (TAD) score looping basal (B) suprabasal cells (SB) fractions. detected distinct SB-specific TAD loop formation evolutionary conserved differentiation complex (EDC) region coincided SE sites. Using Dlx3 conditional knock-out models, we elucidate Dlx3-mediated regulation essential for homeostasis.
